FAQs about NeoClone and MonoClonal Antibodies

What happened to your catalog antibodies like RNA Polymerase Beta, HE4 and GFAP?

BioLegend has acquired NeoClone's legacy antibody portfolio. These products are now only available through BioLegend. NeoClone is still in the Custom monoclonal antibody business and we have expanded our capabilities and services to include Antibody Phage Display (ScFv) and Camelid (VHH) antibodies. Please contact us if you need a new antibody that you just can't find out in the marketplace.

What is a monoclonal antibody?

A monoclonal antibody is a specific antibody usually produced by hybridoma technology. A hybridoma is a fusion of a single antibody producing murine B cell with a myeloma cell to immortalize the antibody producing cell line. A monoclonal antibody recognizes a single epitope of its target protein, making it a very useful tool in research, diagnostics, and therapeutics. To produce a monoclonal antibody, a single hybridoma that produces the specific antibody of interest is isolated and expanded in culture. The antibody is then purified for use. If you'd like a general overview of this topic please check out these pages on antibodies and monoclonal antibodies.

What is ascites?

Monoclonal antibody production can also be done by in vivo methods, which produces an ascites. The in vivo method involves injection of hybridoma cells into the peritoneum of mice where hybridoma cells grow and secrete the antibody of interest. Due to inflammation, ascites fluid is produced and collects in the peritoneum. The ascites fluid contains the antibody produced by the hybridoma cells as well as many other proteins, including background antibodies produced by the mice and albumin.

Antibody concentrations in ascites are typically high, but an accurate concentration of this antibody cannot be readily determined. Ascites is a complex solution containing multiple proteins, background antibodies, as well as the antibody of interest.

How are NeoAbs purified?

Monoclonal antibodies may be purified by various methods. The most common methods are protein A or protein G affinity chromatography. Most sub-classes of antibody have a high affinity for protein A and protein G, thus the antibody binds to these proteins on a resin and extraneous proteins can be washed away. The purified antibody can then be eluted off the resin.

Alternatively, antibodies can be affinity purified utilizing their target immunogen. This method can be expensive and relies on immunogen availability.

NeoClone typically utilizes protein A for purifying antibodies produced by in vitro technology. On occasion, when antibodies need to be purified from ascites, protein G is used.

NeoAb Quality Control

NeoAbs undergo rigorous quality control testing to ensure you receive the highest quality antibodies.  Our tests include one or more of the tested applications and purity analysis by SDS-PAGE.  If you have any questions about quality control of a specific product, please contact one of NeoClone's technical support scientists at 608.260.8190.

Does this antibody recognize the organism I'm researching?

NeoClone posts information on antibody specificity and cross reactivity on our web site and in our technical data sheets. The data comes from sources that include NeoClone's in-house quality control assays, literature reviews and customer feedback.

We believe the results reported in the literature and unpublished feedback are accurate, but do not attempt to verify these results. NeoClone cannot guarantee that your lab can reproduce results reported by other the labs.

If the organism of interest is not referenced on our web site or technical data sheet, NeoClone recommends a literature search, and if the protein sequence is known an alignment comparison. In general, the more conserved proteins are between species, the more likely the antibody will cross react. If the epitope is known, an alignment can determine if the epitope sequence is conserved in a similar region of the protein.

Does this antibody work in my application?

NeoClone posts information about antibody applications on our web site and in our technical data sheets.  The data comes from sources that include NeoClone's in-house quality control assays, literature reviews and customer feedback.  We believe the results reported in the literature and unpublished feedback are accurate, but do not attempt to confirm these results.

If the application of interest is not referenced on our web site or technical data sheet, NeoClone recommends a literature search, to determine if anyone has used that product in such an application.  If you have any questions about applications and NeoAbs do not hesitate to call and ask a NeoClone technical support scientist.

May I have free sample to test in my application?

NeoClone does not provide free samples. Our products are known to work in tested applications and undergo rigorous QC prior to release.
